Size exclusion chromatography (SEC) separates molecules according to their different sizes. In protein separations, this technique is most often used in the intermediate or last stage polishing of purification schemes. It is also an ideal analytical tool for verifying the purity of samples with respect to potential contaminants including aggregated protein.
BioFox SEC resins are produced from agarose beads using a proprietary cross-linking method that results in a highly porous and physically stable agarose matrix. These resins are stable at pressures up to 580 psi allowing for high resolution biochromatography. Two different particle sizes are available for analytical and preparative purposes: BioFox 17 µm; SEC and BioFox 40 µm SEC, respectively. Three different exclusion limits are available: 10,000, 1,200, and 150 kDa.
Agarose-based chromatography supports have been successfully used in for biotechnology research and industrial scale protein purification for decades. Agarose has proven to be exceptionally compatible with naturally occurring biomolecules, most notably proteins, DNA and carbohydrates. The Biofox resin material exhibits negligible non-specific interactions due to the hydrophilic nature of the agarose and is highly chemically resistant. This enables the use of both non-denaturing and denaturing mobile phases. Further, unlike resins made from synthetic polymers, agarose does not have micropores that can contribute to local pH variations in the micro-environment within the column which can contribute to distorted separations.

Resins are resistant to 100% methanol, 100% ethanol, 8 M urea, 6 M guanidine HCl, 30% acetonitrile,
70% formic acid, and 30% trifluoroacetic acid and stable at operating temperatures from 4 to 40°C. |
